Mild & transient: constipation, nausea, vomiting, dizziness, somnolence, bronchospasm, skin irritation, respiratory depression. Sleepy. Risk of drug overdose when drug overdose, withdrawal syndrome when stopping the drug suddenly.
After oral administration, the elimination half-life is 2-4 hours, the cough suppressant effect appears within 1-2 hours and may persist for 4-6 hours. Codeine is metabolised in the liver and eliminated by the kidneys as either free or conjugated with glucuronic acid. Codeine or its metabolites are excreted in the feces very little. Codeine crosses the placenta and small amounts cross the blood-brain barrier.
Codeine is a methylmorphine that replaces the position of hydrogen in the hydroxyl group associated with the aromatic nucleus in the morphine molecule, so codeine has analgesic and cough-reducing effects. However, codeine is better absorbed after oral administration, causing less constipation and less bile spasm than morphine. At therapeutic doses, it causes less bile constriction than morphine) and is less addictive than morphine.Codeine is effective in reducing pain in mild to moderate pain (the analgesic effect of codeine can be attributed to a conversion of about 10% of the dose administered to morphine). Because it causes a lot of constipation if used for a long time, it is recommended to use codeine in combination with anti-inflammatory drugs, non-steroidal anti-inflammatory drugs to increase pain relief and relieve constipation. Coughing center in the medulla oblongata, codeine dries up respiratory secretions and increases the viscosity of bronchial secretions. Codeine is not effective enough to relieve severe cough. Codeine is a cough suppressant in case of dry cough causing insomnia. Codein causes a decrease in intestinal motility, so it is a very good drug in the treatment of diarrhea caused by diabetic neuropathy. Not indicated for acute diarrhea and diarrhea due to infection.
